چکیده

Fibrinolytic therapy progressed by the evolution of different strategies that helped in enhancing its efficacy and specificity. The use microbial fibrinolytic enzymes is leading to a promising direction for treatment cardiovascular diseases. Subtilisin, member subtilases enzyme abundantly found Bacillus species. isolation subtilisin gene (quaking homolog) from subtilis was attempted present work. genomic deoxyribonucleic acid extraction done following Yamada protocol used as template polymerase chain reaction amplification target using specifically designed primers. product ligated into cloning vector pTZ57R/T followed transformation Escherichia coli top 10 strain. A 1090 base pair, partial sequence amplified coding protein 363 amino acids with molecular weight 37550.7 Daltons. nucleotide revealed significant evolutionary relationships other strains subtilis. Our study confirms presence local species suggesting economical way produce important thrombolytic agents commercial pharmaceutical worth.

Bacillus subtilis Subtilisin Gene (aprE) Is Expressed from a crA

In vitro studies demonstrated that the Bacilus subtilis subtilisin gene (aprE) could be transcribed by RNA polymerase holoenzyme reconstituted from core and eA factor obtained from vegetative cells. Upstream deletions (from -45) reduced the amount of transcription from the promoter. A deletion downstream of the promoter that overlapped a putative downstream minor promoter did not affect transcr...

Cloning and Sequence Analysis of Gene Encoding OipA from Iranian Clinical Helicobacter pylori

Background: Outer inflammatory protein A (OipA) is one of the important adhesins of H. pylori and a valuable candidate for vaccine development. Its gene is under "on-off" switch status which correlates with OipA protein expression. Objectives: We aimed to obtain a recombinant OipA clone (with "on" status) from an Iranian clinical isolate. Materials and Methods: A clinical H. pylori-isolate demo...

Isolation, Purification and Characterization of a Thermophilic Alkaline Protease from Bacillus subtilis BP-36

The goal of this research was to isolate and identify the thermostable alkaline protease producing bacteria among several native Iranian microorganisms. At the end of screening program, a Bacillus subtilis BP-36 strain producing thermophilic alkaline protease was isolated from a hot spring in Ardebil province. The target enzyme was purified using a one-step Aqueous two-phase systems (ATPS) prot...

Potent fibrinolytic enzyme from a mutant of Bacillus subtilis IMR-NK1.

A mutant of Bacillus subtilis IMR-NK1, which is used for the production of domestic "natto" in Taiwan, produced high fibrinolytic enzyme activity by solid-state fermentation using wheat bran as medium. In addition, a strong fibrinolytic enzyme was purified from the cultivation media. The purified enzyme was almost homogeneous, as examined by SDS-PAGE and capillary electrophoresis. The enzyme ha...
